New study on NMN

Highlights

Declining NAD(P)H is associated with oocyte dysfunction during reproductive aging


Oocyte quality and fertility can be restored by NMN treatment in aged mice


Supplementation of embryo media with NMN improves developmental milestones


SIRT2 overexpression mimics benefits of NMN but is unlikely to mediate its effects

Treatment with the NAD+ metabolic precursor nicotinamide mononucleotide (NMN) rejuvenates oocyte quality in aged animals, leading to restoration in fertility, and this can be recapitulated by transgenic overexpression of the NAD+-dependent deacylase SIRT2, though deletion of this enzyme does not impair oocyte quality. These benefits of NMN extend to the developing embryo, where supplementation reverses the adverse effect of maternal age on developmental milestones. These findings suggest that late-life restoration of NAD+ levels represents an opportunity to rescue female reproductive function in mammals.

Super interesting is that in the study, the lower dose (.5g/L) worked while the higher dose (1g/L) did nothing, maybe even hindered a little. They mention that maybe there is a point where too much NMN is metabolizing into regular nicotinamide which is a SIRT inhibitor.

Worth taking into consideration here. Looks like more NMN is not always better, at least beyond some point.

Anecdotally, do you know of any menopausal or post menopausal human females who have taken the molecules and had their fertility restored or any that may have actually gone on to fall pregnant and have a successful live birth?

so are there any studies with humans? Or perhaps participants willing to share their experience to create a data set?

this is something that seems logical if NMN works at all that it could restore fertility.
Taking:
Resveratrol 400mg since June 2019; 600mg since October 1
NMN Sublingual 250mg since june 2019 500mg since September 2019; 600mg since Dec 1, 2019
NAD+ Nasal, appx 100mg per day since September 2019; 200mg per day beginning mid November

One forum member has previously posted about a positive result after taking the molecules..

Heard Dr. David Sinclair mentioned that once.

I would agree that this is actually a likely possibility. The only question is how long would one need to be taking the NMN...

But if it becomes a part of a treatment for fertility i imagine the doses would be high for a short term.

Mice have a ridiculously fast metabolism, so i wouldn't know how to translate their results to human.

Exciting as hell though!

Yes mice apparently have to live their whole life in under 24 months (approximately) so it would seem that any noted benefits would occur at a faster speed when compared to humans..
